研究概要

エピジェネティクス機構は、刺激の受容で行われる短期応答、そして、刺激が消失した後にも刺激を受容した記憶を維持する長期応答を通して、遺伝子制御に働いている。他方、転写不活性なクロマチンに位置する遺伝子は、刺激に対して不応になり得る。この転写環境としてのエピゲノム制御には、DNAメチル化とクロマチンの形成が関わり、代謝恒常性の維持と破綻に重要な役割を果たしているが、その機序には不明な点が多い。

本研究では、代謝恒常性に関わるエピジェネティクス機構を理解するために、FAD (flavin adenine dinucleotide)依存性のリジン脱メチル化酵素LSD1ファミリーがエネルギー代謝調節に重要な役割を果たすことを解明する。LSD1およびクロマチン因子に着目し、転写環境とエネルギー代謝のクロストーク制御について解析を行う。具体的には、リジンの脱メチル化による代謝調節とクロマチン変換機構、生命素子としてのFADおよび細胞内FAD生合成経路の役割、DNAとリジンのメチル化による代謝調節とクロマチン変換機構について、分子・細胞・マウス等を用いて解析する。連携研究者の日野と協力して、代謝エピジェネティクスの研究を推進する。

研究概要

エネルギー状態に依存してエピゲノム情報を実際に形成する因子や、エネルギー状態に応じて生成される代謝産物をリガンドもしくは補助因子として利用する因子に着目し、立体構造解析を通してこれらの因子の実像に原子レベルで迫る。手法としてはX線結晶構造解析に加え、溶液散乱曲線から直接構造モデルを構築することができるX線溶液散乱法を用いる。

細胞内の最大のエネルギー消費過程であるリボソームでのタンパク合成をエピジェネティックに制御するNML (Nucleomethylin) はヒストンメチル化酵素Suv39H1、脱アセチル化酵素SIRT1、をリクルートしエピゲノム情報形成因子として働く。一方ガン抑制遺伝子p53はアセチル化酵素であるp300によってアセチル化を受けるが、核小体タンパク質Mybbp1aはこのアセチル化を促進させる。いずれの系も生命素子を利用して転写環境を整えており、これらの分子実体を原子レベルで解明し、代謝とクロストークするエピゲノム情報形成因子の実像と制御機構を構造面から解明する。

代謝シグナルが投射されるゲノム領域の同定と転写環境調節機構の解明

代謝シグナルを受けた遺伝子発現制御機構の中で、転写因子が特定のゲノムDNA配列を読み取る「目」となって作用するには、ゲノムの特定の場所で複数の核内因子と複合体を構成することが重要である。

研究代表者らは、エネルギー代謝シグナルの解析のための個体を用いたアッセイ系として、肝臓へのアデノウイルスによるルシフェラーゼレポーター遺伝子の導入と生体イメージング(IVIS)を組み合わせた定量系 (in vivo Ad-luc解析法)を確立してきた。さらに最近我々は、ゲノム上の全転写因子を網羅する発現ライブラリー (TFEL:Transcription Factor Expression Library) を独自に開発し、それを用いた転写複合体解析法(TFEL scan法)を確立した。

本研究では、in vivo Ad-luc解析法により、様々なエネルギー代謝シグナルが投射されるゲノム上の領域を同定を進め、そこに我々独自のTFEL scan転写複合体解析法を組み合わせることにより、その領域上におけるエピゲノム情報を含めた転写環境制御機構の解明を目指す。

転写環境制御による代謝応答と酸化ストレス応答のクロストーク

細胞の代謝様式は、静止期と増殖期では大きく異なる。静止期では定常状態の維持が重要であり、酸素呼吸が生体の構成因子を常に酸化障害の危険に曝していることを鑑みれば、酸化ストレスからの防御が最も基本的な恒常性維持機構である。一方、増殖期では、核酸、細胞膜、細胞内小器官など細胞構成因子を新規に合成する必要があり、細胞はグルコースやグルタミンを大量に取り込み、同化反応を活性化させている。増殖シグナルは、細胞の代謝様式を改変すること(代謝リプログラミング)により、細胞の増殖を可能にしている。最近、我々は酸化ストレス応答の鍵因子である転写因子Nrf2が、増殖シグナルにより機能変換を受けて代謝リプログラミングを推進することを見いだした。

そこで、本研究では、代謝リプログラミングを支える分子機構をさらに明らかにするために、Nrf2による同化反応制御機構の解析と代謝物によるNrf2機能制御機構の解析を行う。増殖シグナルによるNrf2の標的遺伝子プロファイルの変化、Nrf2複合体の変化、標的遺伝子近傍のエピゲノム環境の変化、さらに代謝物の変化を網羅的に検討して、細胞増殖を促進する代謝リプログラミングの実態とそれを支える分子機構に迫る。また、がん細胞特有の代謝物プロファイルがNrf2の機能発現に影響を及ぼしている可能性を見いだしているので、それらがNrf2複合体やエピゲノム環境に及ぼす影響を検討する。これにより、がん細胞における代謝と転写環境のダイナミックな相互作用を明らかにし、その悪性化や治療抵抗性との関係を解明する。

研究概要

エネルギー代謝や環境要因によって日常的に引き起こされるDNA塩基損傷の発生とその修復は、それ自体が局所的なエピゲノム環境の影響を受けると同時に、あらかじめ書き込まれたエピゲノム情報を攪乱し、書き換える (“Rewriting”) 機会を与えるものと捉えることができるがその詳細な分子機構については不明な点が多く残されている。本研究ではゲノム配列情報の維持にあたるDNA修復機構と、エピゲノム情報の維持・再編に関わる分子機構との相互制御関係(クロスレギュレーション)を分子レベルで明らかにすると共に、その破綻によって引き起こされるゲノム不安定性やエピゲノム異常と種々の疾患・病態発現機構との関連を理解することを目的とする。具体的には、以下のような問いに答えることを目指して研究を進める。

研究概要

核内におけるDNAやヒストンなどのメチル化反応は、S-adenosyl-L-methoinine (SAM)をメチル基供与体とする。SAMはメチオニンとATPを基質としてmethionine adenosyltransferase (MAT) により合成される。そのアイソザイムMATIIはメチル化酵素やクロマチン構造制御因子と共に複合体(SAMITと命名)を形成し、転写因子により特定の標的遺伝子へ動員され、周辺ヒストンのメチル化を促進する。

そこで本計画研究では、細胞分化に伴うエピゲノムリモデリングの優れたモデルとなるBリンパ球-形質細胞分化を取りあげ、SAMIT複合体の機能、構造、生物・医学的意義を明らかにする。そして、メチオニン代謝回路とエピゲノムの共役機構を理解する。具体的には、MATIIの核局在機構や、ChIP-Seq を用いたMATIIのクロマチン結合部位およびヒストン修飾のマッピングとその分化段階に応じた変動の検討を進める。また、分化段階に応じたSAMIT複合体組成の変動も調べる。並行して、Bリンパ球でのノックダウンや遺伝子改変マウス作製などにより、MATIIおよびSAMIT複合体構成因子の液性免疫における機能を調べる。

DNA及びタンパク質のリジン残基とアルギニン残基へのメチル化は転写環境の構築に不可欠であるが、メチル基供与体・S-adenosyl-L-methionine (SAM)は、メチオニンとATPからSAM合成酵素 (SAMS、またはmethionine adenosyltrasferase, MAT) によって生体内で生成される。このように、転写環境の整備とアミノ酸代謝経路には密接なリンクがあるが、そのクロストーク制御は全く不明である。

そこで本研究は、転写環境に及ぼすメチル基転移の根幹的重要性を理解するために、SAM代謝系との相互作用について線虫 (C. elegens) の4種類のsams変異体 (sams-1、sams-3〜5) を用いて、遺伝学・生化学的解析から基本的な生命機能を解明する。メチル化酵素はゲノム中に1%程度コードされており、線虫には116個の遺伝子が存在する。具体的には、これら全てをRNAi法でノックダウンしてsams機能と直結するメチル化酵素を探索し、遺伝学・生化学的に解析する。さらに、研究分担者の高橋と協力して、メチオニン以外の他のアミノ酸代謝系にも着目し、マウスなどのモデル動物も活用し、転写環境の構築とのクロストーク制御を明らかにする。
